Several factors are contributing to an increased air of excitement about the eukaryotic DNA replication problem: new insights into the nature of origins of replication, a better appreciation of the factors that control initiation, and studies of a DNA polymerase α‐primase enzyme complex. In this review, recent research on the initiation, elongation and termination phases of DNA replication is critically examined and a coherent picture is formulated. In the not‐far‐distant future we expect to reproduce these processes in biochemically defined systems.

The article begins with an effort to clarify and differentiate a variety of terms used by analysts in dealing with mentions of persons in conversation and other forms of talk-in-interaction — such terms as person-reference, identifying, describing, categorizing, and the like. This effort leads to the observation that `reference to persons' and `membership categorization' are quite distinct sets of practices, with most reference to persons not being done by membership categories, and most uses of membership categorization devices being in the (...) service of actions other than referring. Two interactional sequences whose analysis turns on a connection to talk earlier in the occasion are then examined; first, to establish what is going on interactionally without respect to the mentioning of persons, and then as exercises in examining the various ways person-reference and membership categorization can figure in a stretch of interaction. In a period given to emphasizing diversity among humans, we would do well to explore diversity among forms of discourse and among forms of talk-in-interaction in particular. Among the speech-exchange systems, ordinary conversation has been claimed to be distinctive and fundamental, but questions have been raised about both claims. The resources for discriminating among speech-exchange systems are located in such generic organizations of practice as turn-taking, sequence organization, the organization of repair and the overall structural organization of episodes of interaction. I try to show that `conversation' as a distinctive speech-exchange system is real and is not only a residual category, and that it is to be understood as the `basic' speech-exchange system, in part by reference to the distinctive turn-taking organization through which it is implemented. The `motivation' for having developed a formal account of this turn-taking organization is recounted, and that formal account is defended for its usability in the analytic explication of singular, contexted episodes of talk. The remainder of the article is given over to such an exemplary account - an examination of an episode of interaction during a testing session between a man whose brain hemispheres had been surgically separated and a researcher. The article begins with a sketch of the relation of interaction to language and to culture, and of the students of interaction to the students of language and of culture. A 10-second segment of recorded interaction at a family dinner is then examined in a fashion meant to preserve the integrity1 of what is being done interactionally while incorporating attention to the deployment of various facets of the language that is used, and its relationship to simultaneously ongoing bodily doings. An (...) interactional practice – whining – from that episode is then juxtaposed with the same practice in several other segments of interaction in the interests of developing a more formal, transsituational account. The viability of research focused on phenomena in an analytically distinct domain of events while preserving the integrity of the occasions in which instances of the phenomenon occurred is then reviewed, using a case study of the conjoint use of phonetic analysis and conversation analysis. The article concludes with a reply to Levinson's article in this special issue of the journal, and uses the occasion to sketch the relationship between interaction and so-called ‘macro’ social and cultural formations such as kinship. Turns-at-talk are fundamental units of participation in talk-in-interaction, and turn-constructional-units are the basic building blocks for turns. Possible completion of a TCU is, in principle, the possible completion of the turn, but multi-unit turns are not uncommon, and participants have practices for constructing multi-unit turns and for recognizing them in the course of their production. This article offers an account of one practice usable by speakers and recipients to convey and recognize the designed completion of a multi-TCU turn and/or a (...) multi-turn sequence in which ‘answering’ is being done: returning to, or ‘re-using’, a word or phrase from the start of the turn or sequence, whether articulated by same or different speaker, whether used to refer to same or different referents. This practice is one of the resources by which the overall structural organization of an interactional unit and its local realization are mutually realized. (shrink)

Although there is no lack of reasons for conversation analysis to be reluctant to adopt a cognitivist idiom and paradigm in studying talk and other conduct in interaction, examination of the literature with an open mind will disclose attentiveness to such themes in the conversation-analytic literature nonetheless. The pursuit of such themes however, cannot be appropriately and successfully conducted under the aegis of currently dominant cognitivist paradigms. One central analytic resource in CA work is the notion of a ‘possible X’, (...) a resource which is here described and exemplified for three discrete ‘values’ of ‘X’. The understanding of how such ‘possible Xs’ could work for participants in interaction invites understanding by analysts by reference to a ‘multiple passes’ model of uptake, a characterization which for now can be no more than metaphoric. Here is a venue at which conversation-analysts and neuro/cognitive analysts might usefully try to work together. (shrink)

One important lesson of Roberts' target article may be potentially obscured for some by the title's reference to “self-experimentation.” At the core of this work, the key investigative resource is sustained and systematic observation, not experimentation, and it is deployed in a fashion not necessarily restricted to self-examination. There is an important reminder here of a strategically important, but neglected, relationship between observation and experiment.

Although Harvey Sacks' `Max Weber's Ancient Judaism' is an early student paper, it raises issues of theory, method and disciplinary mandate which have continuing relevance. I frame the article in two ways. First, I sketch the academic and intellectual context in which the paper was written, in particular the institutional setting in Berkeley of the early 1960s, and the activities and preoccupations animating the work of the group of students which was the most proximate context for Sacks' writing at this (...) time, including the efforts to come to terms with Garfinkel's early work. Second, I relate this article to two others written by Sacks in roughly the same period, and sketch a thematic continuity and development running through them - a theme whose endpoint is the centrality of what must surely be called `culture' to the calling of Sociology. I share the authors' stance on the dialogic or interactional character of language. The authors, however, have left actual interaction out of their conception of dialogue. I sketch a number of organizations of practices of talking and understanding that supply the basic arena for talk-in-interaction. It is by reference to these that mechanisms for speech production and understanding need to be understood.

Because rate of force development is an emerging outcome measure for the assessment of neuromuscular function in unfatigued conditions, and it represents a valid alternative/complement to the classical evaluation of pure maximal strength, this scoping review aimed to map the available evidence regarding RFD as an indicator of neuromuscular fatigue. Thus, following a general overview of the main studies published on this topic, we arbitrarily compared the amount of neuromuscular fatigue between the “gold standard” measure and peak, early and late (...) RFD. Seventy full-text articles were included in the review. The most-common fatiguing exercises were resistance exercises, endurance exercises/locomotor activities, isokinetic contractions, and simulated/real sport situations. The most widely tested tasks were knee extension and plantar flexion. The reason for evaluating RFD was lacking in 36% of the studies. On average, the amount of fatigue for MVF was comparable to late RFD but lower compared to both peak RFD and early RFD. Even if the rationale for evaluating RFD in the fatigued state was often lacking and the specificity between test task and fatiguing exercise characteristics was not always respected in the included studies, RFD seems to be a valid indicator of neuromuscular fatigue. Based on our arbitrary analyses, peak RFD and early phase RFD appear even to be more sensitive to quantify neuromuscular fatigue than MVF and late phase RFD. (shrink)

In several recent papers and a monograph, Andreas Stokke argues that questions can be misleading, but that they cannot be lies. The aim of this paper is to show that ordinary speakers disagree. We show that ordinary speakers judge certain kinds of insincere questions to be lies, namely questions carrying a believed-false presupposition the speaker intends to convey. These judgements are robust and remain so when the participants are given the possibility of classifying the utterances as misleading or as deceiving. (...) The judgements contrast with judgements participants give about cases of misleading or deceptive behaviour, and they pattern with judgements participants make about declarative lies. Finally, the possibility of lying with non-declaratives is not confined to questions: ordinary speakers also judge utterances of imperative, exclamative and optative sentences carrying believed-false presuppositions to be lies. In his famous essay "The Ethics of Belief," William K. Clifford claimed "it is wrong always, everywhere, and for anyone, to believe anything upon insufficient evidence." ). One might claim that a corollary to Clifford's Law is that it is wrong, always, everywhere, and for anyone, to withhold belief when faced with sufficient evidence. Seeming to operate on this principle, many religious philosophers—from St. Anselm to Alvin Plantinga—have claimed that non-believers are psychologically or cognitively deficient if they refuse to believe (...) in the existence of God, when presented with evidence for His existence in the form of relevant experience or religious arguments that are prima facie unassailable. Similarly, many atheists fail to see how believers can confront the problem of evil and still assert their belief in a benevolent, omnipotent, and omniscient Creator. In this paper, I propose to explain why religious arguments so often fail to persuade. In doing so, I first offer an account of persuasion and then apply it to religious arguments. I go on to argue that at least some religious arguments commit a form of question-begging, which I call "begging the doxastic question." An argument begs the doxastic question, on my account, when a subject would find the argument persuasive only if she antecedently believes the argument's conclusion. This form of question begging is not, strictly speaking, a case of circularity and thus, is not a fallacy; rather, it would explain why one coming to the argument would fail to be persuaded by it unless he already accepted its conclusion. This has the effect, when applied to religious argumentation, that religious arguments are rarely persuasive, which raises the further question: what good are religious arguments? I end by suggesting some non-persuasive functions of religious argument. Finally, I suggest that a full understanding of religious argumentation should give evidentialists pause, for religious beliefs look less like belief states that are sensitive to evidentiary states and more like framework principles or fundamental commitments. Drawing on evolutionary epistemology, process ontology, and a social-cognition approach, this book suggests cognitive evolution, an evolutionary-constructivist social and normative theory of change and stability of international social orders. It argues that practices and their background knowledge survive preferentially, communities of practice serve as their vehicle, and social orders evolve. As an evolutionary theory of world ordering, which does not borrow from the natural sciences, it explains why certain configurations of practices organize and govern social orders epistemically and normatively, and (...) why and how these configurations evolve from one social order to another. Suggesting a multiple and overlapping international social orders' approach, the book uses three running cases of contested orders - Europe's contemporary social order, the cyberspace order, and the corporate order - to illustrate the theory. Based on the concepts of common humanity and epistemological security, the author also submits a normative theory of better practices and of bounded progress. (shrink)

Der zweite Band sollte die "Philosophie des kontemplativen Lebens," der dritte die des "aktiven Lebens" behandeln. Der Umriß des Systems, der im ersten Bande mitunter vielleicht noch wie ein leeres Rahmen- und Fächerwerk wirken könnte, sollte nun seine (...) Brauchbarkeit zeigen. Er sollte angefüllt und bereichert werden durch sachhaltige Untersuchungen über die einzelnen Teilgebiete des philosophischen Systems. So war für den zweiten Band z. B. eine Wissenschaftslehre (Logik und Methodologie), eine Philosophie der Kunst (Ästhetik) und eine Behandlung der mystischen Religiosität geplant. The distinction between lying and mere misleading is commonly tied to the distinction between saying and conversationally implicating. Many definitions of lying are based on the idea that liars say something they believe to be false, while misleaders put forward a believed-false conversational implicature. The aim of this paper is to motivate, spell out, and defend an alternative approach, on which lying and misleading differ in terms of commitment: liars, but not misleaders, commit themselves to something they believe to be (...) false. This approach entails that lying and misleading involve speech-acts of different force. While lying requires the committal speech-act of asserting, misleading involves the non-committal speech-act of suggesting. The approach leads to a broader definition of lying that can account for lies that are told while speaking non-literally or with the help of presuppositions, and it allows for a parallel definition of misleading, which so far is lacking in the debate. With the world grappling with continued spread of monkeypox internationally, vaccines play a crucial role in mitigating the harms from infection and preventing spread. However, countries with the greatest need - particularly historically endemic countries with the highest monkeypox case-fatality rates - are not able to acquire scarce vaccines. This is unjust, and requires rectification through equitable allocation of vaccines globally. We propose applying the Fair Priority Model for such allocation, which emphasizes three key principles: 1) preventing harm; 2) prioritizing (...) the disadvantaged; and 3) treating people with equal moral concern. Post-exposure prophylaxis (PEPV) has the most potential to mitigate harm, and so ensuring countries have sufficient supply for PEPV should be the first priority. And historically endemic countries, which face disadvantages that compound potential harms from monkeypox, should be the first recipients of such vaccines. Once sufficient supply is allocated for countries to apply PEPV, global allocation could move on to pre-exposure prophylaxis (PrEP), again prioritizing historically endemic countries first before distribution to the rest of the global community, based on projected number of cases and vulnerability to harm. Background In the course of the COVID-19 pandemic, the biomedical research community’s attempt to focus the attention on fighting COVID-19, led to several challenges within the field of research ethics. However, we know little about the practical relevance of these challenges for Research Ethics Committees. Methods We conducted a qualitative survey across all 52 German RECs on the challenges and potential solutions with reviewing proposals for COVID-19 studies. We de-identified the answers and applied thematic text analysis for the extraction and (...) synthesis of challenges and potential solutions that we grouped under established principles for clinical research ethics. Results We received an overall response rate of 42%. The 22 responding RECs reported that they had assessed a total of 441 study proposals on COVID-19 until 21 April 2020. For the review of these proposals the RECs indicated a broad spectrum of challenges regarding social value, scientific validity, favourable risk–benefit ratio, informed consent, independent review, fair selection of trial participants, and respect for study participants. Mentioned solutions ranged from improved local/national coordination, over guidance on modified consent procedures, to priority setting across clinical studies. Conclusions RECs are facing a broad spectrum of pressing challenges in reviewing COVID-19 studies. Some challenges for consent procedures are well known from research in intensive care settings but are further aggravated by infection measures. Other challenges such as reviewing several clinical studies at the same time that potentially compete for the recruitment of in-house COVID-19 patients are unique to the current situation. For some of the challenges the proposed solutions in our survey could relatively easy be translated into practice. Others need further conceptual and empirical research. Our findings together with the increasing body of literature on COVID-19 research ethics, and further stakeholder engagement should inform the development of hands-on guidance for researchers, funders, RECs, and further oversight bodies. Health care costs have been rising steadily in most industrialized countries. These increases are driven primarily by technological advances and, to a lesser degree, by aging of the population. Many factors make it unlikely that market forces alone will limit increases in the costs of health care. These unremitting increases make health care rationing appear both necessary and inevitable.One of the least controversial mechanisms for rationing could be to allow patients to make their own choices as to which kinds of (...) care they would be willing to forgo. This is appealing because it preserves individual freedom of choice regarding health care in a way that other rationing mechanisms often do not. Rationing by patient choice, however, can only happen if patients recognize that resources are limited and need to be conserved, and are willing to forgo marginal benefits. (shrink)

It is widely held that all lies are assertions: the traditional definition of lying entails that, in order to lie, speakers have to assert something they believe to be false. It is also widely held that assertion contrasts with presupposition and, in particular, that one cannot assert something by presupposing it. Together, these views imply that speakers cannot lie with presuppositions—a view that Andreas Stokke has recently explicitly defended. The aim of this paper is to argue that speakers can lie (...) with presuppositions, and to discuss some of the implications this outcome has for current research on lying, assertion and presupposition. In Emanuel Adler's distinctive constructivist approach to international relations theory, international practices evolve in tandem with collective knowledge of the material and social worlds. This book - comprising a selection of his journal publications, a new introduction and three previously unpublished articles - points IR constructivism in a novel direction, characterized as 'communitarian'. Adler's synthesis does not herald the end of the nation-state; nor does it suggest that agency is unimportant in international life. Rather, it argues that what mediates (...) between individual and state agency and social structures are communities of practice, which are the wellspring and repositories of collective meanings and social practices. The concept of communities of practice casts new light on epistemic communities and security communities, helping to explain why certain ideas congeal into human practices and others do not, and which social mechanisms can facilitate the emergence of normatively better communities. Molecular biologists exploit information conveyed by mechanistic models for experimental purposes. In this article, I make sense of this aspect of biological practice by developing Keller’s idea of the distinction between ‘models of’ and ‘models for’. ‘Models of (phenomena)’ should be understood as models representing phenomena and are valuable if they explain phenomena. ‘Models for (manipulating phenomena)’ are new types of material manipulations and are important not because of their explanatory force, but because of the interventionist strategies they afford. This (...) is a distinction between aspects of the same model. In molecular biology, models may be treated either as ‘models of’ or as ‘models for’. By analysing the discovery and characterization of restriction–modification systems and their exploitation for DNA cloning and mapping, I identify the differences between treating a model as a ‘model of’ or as a ‘model for’. These lie in the cognitive disposition of the modeller towards the model: a modeller will look at a model as a ‘model of’ if interested in its explanatory force, or as a ‘model for’ if interested in the material manipulations it can possibly afford. In spite of the fact that most university campuses are considered bastions of liberalism (in contrast to the conservative “mainstream”), it remains difficult for lesbian, gay, bisexual, and transgendered faculty members and students to come out, and instances of poor treatment still occur. In addition, curricular issues related to LGBT identities and concerns continue to be met with resistance or outright hostility. In this paper, I argue that all faculty—not just LGBT faculty—have an obligation to play a more active role (...) in creating an LGBT-friendly environment on their respective campuses. To understand the endogenous components of the event-related brain potential (ERP), we must use data about the components' antecedent conditions to form hypotheses about the information-processing function of the underlying brain activity. These hypotheses, in turn, generate testable predictions about the consequences of the component. We review the application of this approach to the analysis of the P300 component. The amplitude of the P300 is controlled multiplicatively by the subjective probability and the task relevance of the eliciting events, whereas its (...) latency depends on the duration of stimulus evaluation. These and other factors suggest that the P300 is a manifestation of activity occurring whenever one's model of the environment must be revised. Tests of three predictions based on this “context updating” model are reviewed. Verleger's critique is based on a misconstrual of the model as well as a partial and misleading reading of the relevant literature. (shrink)

Frequent public uproar over forms of data science that rely on information about people demonstrates the challenges of defining and demonstrating trustworthy digital data research practices. This paper reviews problems of trustworthiness in what we term pervasive data research: scholarship that relies on the rich information generated about people through digital interaction. We highlight the entwined problems of participant unawareness of such research and the relationship of pervasive data research to corporate datafication and surveillance. We suggest a way forward by (...) drawing from the history of a different methodological approach in which researchers have struggled with trustworthy practice: ethnography. To grapple with the colonial legacy of their methods, ethnographers have developed analytic lenses and researcher practices that foreground relations of awareness and power. These lenses are inspiring but also challenging for pervasive data research, given the flattening of contexts inherent in digital data collection. We propose ways that pervasive data researchers can incorporate reflection on awareness and power within their research to support the development of trustworthy data science. (shrink)
